Purification
Services
ICT offers antibody development and purification
services. Antibodies can be purified via Protein
A, Protein G, and affinity purified. Using your
antibody, we can purify the antigen as well. Every
project is performed specifically for each client,
ensuring maximum utility and specificity.
Purification projects may take a few days to a few weeks
to complete.
Purification of antibodies and
antigens enhances their reactivity and utility for
improved results in many applications (i.e.,
immunoassays and flow cytometry). Monoclonal and
polyclonal antibodies can be purified and concentrated
to meet your specific project requirements. Purification
and concentration of IgG can be done using Protein A or
Protein G matrices. Other classes of immunoglobulins
(such as IgA, IgD, IgE, IgM, and IgY), may be isolated
using alternative techniques, as the specific
purification methodology is dependent upon the species
and class of immunoglobulin chosen.
Affinity purification
techniques can be used to purify antibodies by capturing
them on an affinity gel matrix containing covalently
coupled analyte. Purified antigen can also be obtained
by coupling antibodies to an affinity column. When our
clients provide us with the analyte or antigen sample
specific to their research, we can provide the affinity
purified product.
In order to serve you better,
please review and answer as many of the following
questions as best you can. Please print out the
questions and fax us your answers at 952-888-8988 or
simply give us a call at 1-800-829-3194 and we can go
through it together.
Antibody
Purification Questionnaire
1. Is a monoclonal or
polyclonal antibody to be purified?
2. What class, isotype, or subclass of antibody is to be
purified?
3. In what animal was the antibody made (for example,
rabbit)?
4. What matrix is the antibody currently in (for
example, PBS, frozen serum)?
5. What is the composition of the matrix/buffer (such as
pH, salt concentration, etc.)?
6. Has the antibody been manipulated at all (for
example, freeze/thaw cycles)?
7. How much of the antibody will be purified (for
example, 200mL serum)?
8. What is the expected concentration of the antibody?
9. What is the purified antibody to be used for?
10. Will this be an on-going project, or a 1-time
special project?
11. If on-going, how many antibodies will be purified at
one time?
12. Do you require affinity purification?
12a. Do you have an affinity
column?
12b. Is the antigen available
or can it be purchased?
12c. Are there any
cross-reacting antigens that may react with the
antibody?
13. For a monoclonal antibody, is it to be purified from
ascites fluid or a tissue culture system?
14. What type of tissue culture system was used, if
any?
14a. What is the composition of
the supernatant?
14b. How many times has the
supernatant been concentrated?
15. What is the expected volume of ascites or
supernatant to be purified?
16. Have you tried purifying it before?
16a. If so, what methods were
tried and were there any particular problems?
