These kits come in either Green (FAM), or Red (SR-101)
Quantitate serine protease activity in whole living cells without using antibodies. Measure chymotrypsin-like protease activation in whole living cells using ICT’s new FLISP kits. Our FLISP reagents (Fluorescent Labeled Inhibitors of Serine Proteases) are cell-permeant inhibitors linked to an amino acid targeted by serine proteases linked to a fluorescent label, such as FAM (carboxyfluorescein = green) or SR-101 (Sulforhodamine 101 = red).

The FFCK kit detects chymotrypsin-like serine proteases that favor phenylalanine

FLCK
The FLCK kit is for those that favor leucine; use the kits with an additional spacer sequence to reduce steric hindrance at the binding site of the serine protease.

Sample protocol:
- Culture your cells.
- Subject cells to your experimental protocol.
- Reconstitute the reagent to form the stock concentrate.
- Dilute the stock concentrate to the final working solution.
- Add the working solution directly to the cell culture for labeling.
- If desired, concurrently label cells with DAPI, or SR-VAD-FMK.
- Incubate for 1-4 hours.
- Wash cells.
- If desired, fix cells.
- Analyze data with a fluorescence microscope, fluorescence plate reader, or flow cytometer.
