TMB Super Sensitive 1-Component HRP Microwell Substrate (SUBS) is a ready-to-use TMB substrate for microwell applications containing 3,3′,5,5′-tetramethylbenzidine in a mildly acidic buffer. 3,3′,5,5′-tetramethylbenzidine (TMB) substrates react with peroxidase, yielding a soluble blue reaction product. When the TMB-peroxidase reaction is stopped using Stop Solution for TMB Substrates, the chromogen changes from blue to yellow, and the absorbance changes from 620 nm to 450 nm. TMB Super Sensitive HRP Microwell Substrate is our most sensitive TMB buffer. This sensitive, faster kinetic TMB substrate is approximately 40-fold more sensitive than our most popular substrate, TMB 1-Component HRP Microwell Substrate. This product is ideal for detecting very low levels of a target molecule, running samples that must be highly diluted (1:10,000), amplifying the signal when using antibodies with low binding capacity, running samples that exhibit high steric hindrance, and shortening the incubation time of the assay.   1. Allow product to equilibrate to room temperature (25°C) prior to use.
  2. Add 100 µL to each well. When reacting with peroxidase, a soluble blue reaction product develops that can be read at 370 nm or in a range of 620-650 nm. The reaction product is stable for 1 hour.
  3. The reaction can be stopped using an appropriate stop solution, such as Stop Solution for TMB Substrates (STOPT), for endpoint assays. STOPT changes the chromogen from blue to yellow and the absorbance to 450 nm. For best results, sample absorbance should be monitored and read before values exceed 2.0 OD units. STOPT can increase the absorbance up to 3-fold, so the reaction should be stopped when absorbance values reach approximately 0.7 OD units.
